It is located on Piazza San Pietro. I can give you the exact schedule because I have it in my agenda... Mon-Fri (from 8.30am to 7pm) and Sat (from 8.30am to 6pm), Sunday - closed. If you just have letters to send, there are the blue Vatican post boxes (pay attention to put the proper Vatican stamp).
